Many companies offer automation for various sample preparation processes. Perhaps the most common question we receive at iChrom Solutions is, “What makes on-line SPE so special? The answer has a few main parts:
- SPE in general is widely-accepted as the highest-selectivity technique for maximum sample cleanup (i.e., resulting in the purest extracts with the fewest interferences). Because it is a chromatographic technique, SPE can take advantage of slight molecular differences in sample components to isolate them from one another, unlike liquid/liquid extraction or protein precipitation.
- Because on-line SPE cartridges (CHROSPE) are processed under pressure (like HPLC columns), it is possible to use microparticulate packings, which means a) higher-efficiency elutions with smaller volumes, b) higher capture efficiencies, requiring less sorbent than off-line approaches, c) compatibility for direct elution from the extraction cartridge into the analytical column. This means zero analyte loss due to dry-downs, reconstitutions, vessel transfers, etc.
- Our latest on-line SPE technology is even compatible with UHPLC analyses, allowing cartridge elutions at up to 1,000 bar (~15,000 psi). Try doing that with an off-line column!
- Full automation. In many cases the user simply places the raw liquid sample into the sample-handler vial, and pushes “go”. The system takes it from there, from further raw sample processing (e.g., internal standard addition, dilute and mix, etc.), to complete extraction protocols on the high-performance cartridge, through the analytical process. All with integrated control via CHRONOS software…including interface with the mass spectrometric detector.
UHPLC is the unquestioned leader in advanced liquid chromatography separation techniques. The high-pressures provided by UHPLC pumps (up to ~18,000 psi/~1200 bar) allow for columns with sub-2 micron particles for previously-unseen efficiencies and separation speeds. iChrom Solutions provides complete UHPLC systems with advanced pumps and highly-configurable, flexible autosamplers to perform the necessary additional tasks needed for integration with on-line SPE modules (e.g., handling not only finished, but raw samples).
